    Cultivated peanut (Arachis hypogaea L.) forms root nodules to enable a symbiotic relationship with rhizobia for biological nitrogen fixation. To understand the genetic factors of peanut nodulation, it is fundamental to genetically map and clone the genes involved in nodulation. For genetic mapping, high throughput genotyping with a large number of polymorphic markers is critical. In this study, two sets of sister recombinant inbred lines (RILs), each containing a nodulating (Nod+) and non-nodulating (Nod-) line, and their Nod+ parental lines were extensively genotyped. Several next generation sequencing (NGS) methods including target enrichment sequencing (TES), RNA-sequencing (RNA-seq), genotyping by sequencing (GBS), and the 48K Axiom Arachis2 SNP array, and various analysis pipelines were applied to identify single nucleotide polymorphisms (SNP) among the two sets of RILs and their parents. TES revealed the largest number of homozygous SNPs (15,947) between the original parental lines, followed by the Axiom Arachis2 SNP array (1,887), RNA-seq (1,633), and GBS (312). Among the five SNP analysis pipelines applied, the alignment to A/B genome followed by HAPLOSWEEP revealed the largest number of homozygous SNPs and highest concordance rate (79%) with the array. A total of 222 and 1,200 homozygous SNPs were polymorphic between the Nod+ and Nod- sister RILs and between their parents, respectively. A graphical genotype map of the sister RILs was constructed with these SNPs, which demonstrated the candidate genomic regions harboring genes controlling nodulation across the whole genome. Results of this study mainly provide the pros and cons of NGS and SNP genotyping platforms for genetic mapping in peanut, and also provide potential genetic resources to narrow down the genomic regions controlling peanut nodulation, which would lay the foundation for gene cloning and improvement of nitrogen fixation in peanut. The ability to correctly identify metal ion ligand-binding residues is valuable for the human health and the design of molecular drug. Precisely identifying these residues, however, remains challenging work. We have presented an improved computational approach for predicting the binding residues of 10 metal ion ligands (Zn2+, Cu2+, Fe2+, Fe3+, Co2+, Ca2+, Mg2+, Mn2+, Na+, and K+) by adding reclassified relative solvent accessibility (RSA). The best accuracy of fivefold cross-validation was higher than 77.9%, which was about 16% higher than the previous result on the same dataset. It was found that different reclassification of the RSA information can make different contributions to the identification of specific ligand binding residues. Our study has provided an additional understanding of the effect of the RSA on the identification of metal ion ligand binding residues. Sensor technologies could be applied to obtain objective evaluation of turkey gait. Inertial measurement units (IMUs) measure acceleration and rotational velocity, which makes them attractive devices for gait analysis. The aim of this study was to compare three different methods for step detection from IMU data from turkeys. This is an essential step for future feature extraction for the evaluation of turkey locomotion. Data from turkeys walking through a corridor with IMUs attached to each upper leg were annotated manually. We evaluated change point detection, local extrema approach, and gradient boosting machine in terms of step detection and precision of start and end point of the steps. All three methods were successful in step detection, but local extrema approach showed more false detections. In terms of precision of start and end point of steps, change point detection performed poorly due to significant irregular delay, while gradient boosting machine was most precise. For the allowed distance to the annotated steps of 0.2 s, the precision of gradient boosting machine was 0.81 and the recall was 0.84, which is much better in comparison to the other two methods ( less then 0.61). At an allowed distance of 1 s, performance of the three models was similar. Gradient boosting machine was identified as the most accurate for signal segmentation with a final goal to extract information about turkey gait; however, it requires an annotated training dataset. WFO has been gradually used in China, but limited reports on whether WFO is suitable for Chinese patients. This study aims to investigate the concordance of treatment options between WFO and real clinical practice for Cervical cancer patients retrospectively. We retrospectively enrolled 300 cases of cervical cancer patients. WFO provides treatment options for 246 supported cases. Real clinical practice were defined as concordant if treatment options were designated “recommended” or “for consideration” by WFO. Concordance of treatment option between WFO and real clinical practice was analyzed statistically. The treatment concordance between WFO and real clinical practice occurred in 72.8% (179/246) of cervical cancer cases. Logistic regression analysis showed that rural registration residences, advanced age, poor ECOG performance status, stages II-IV disease have a remarkable impact on consistency. selleck compound The main reasons attributed to the 27.2% (67/246) of the discordant cases were the substitution of nedaplatin for cisplatin, reimbursement plan of bevacizumab, surgical preference, and absence of neoadjuvant/adjuvant chemotherapy and PD-1/PD-L1 antibodies recommendations.
