Discrepancies exist in backpack load recommendations for obese/overweight children, and the recommendations do not consider school trolleys. This study analysed obese/overweight and healthy-weight students’ perceived load and fatigue when carrying schoolbags and their gait kinematics and rate of perceived exertion (RPE) when carrying backpacks or pulling school trolleys with different loads. Twelve obese/overweight and 36 healthy-weight students were asked about their perceived load and fatigue in carrying their schoolbags to school. MS023 Then, a kinematic gait analysis was completed in students walking unloaded or transporting 10%, 15% or 20% of their bodyweight (BW) in a backpack or trolley. RPE was recorded after each condition. The average hip rotation and knee adduction angles differed between body mass index (BMI) groups. The healthy-weight group reported higher RPEs than the overweight/obese group when pulling a trolley with 10-15% BW. In conclusion, both BMI groups responded similarly to load and schoolbag type.Operating a small carry-on unmanned aerial system (UAS) alone is challenging. Research on facilitating single-operator work has focused mainly on payload operation and health monitoring. Little focus has been given to mission-related aspects and how the command and control (C2) map display contributes to mission accomplishment. This study uses cognitive work analysis (CWA) to describe the operational work of the mission operator of a Skylark miniature UAS system. Three CWA phases were conducted – work domain analysis, control task analysis and strategy analysis – providing a rich framework of operational mission phases, task components, processes and the physical interface-objects in use. These representations highlight the operators’ extensive use of the C2 map during all mission phases, for all object-related processes. To further enhance the outcomes of the CWA, and prior to outlining specific design requirements, an empirical investigation was conducted in which the eye movements of five experienced operators were obtained during a simulated mission. The empirical results confirm and further specify the work patterns that operators adopt. Quantitative analysis shows operators’ extensive focus on the map, especially during mission-critical phases. These analyses led to the conclusion that a significant change in the way operators interact with the C2 map, or alternative designs to enhance map-based information utilization, should be applied. Insights drawn from this analysis can be applied to other aerial surveillance work domains, and adding empirical evaluations is helpful to further refine and reinforce the CWA outcomes.Benzo[a]pyrene (BaP) is a hazardous compound for human health and for environmental compartments. Its transfer and deposition through the atmosphere affects soil quality. In this context, we quantified the content of BaP and other Polycyclic Aromatic Hydrocarbons (PAHs) in the soils of a prominent Coal Region in Transition to test whether the soil screening levels in force are realistic and whether they reflect the complexity of regions closely linked to heavy industries and mining. In this regard, soil screening levels are thresholds often established without considering historical anthropogenic activities that affect soil (diffuse pollution). The 150 soil samples studied showed a notable content of high molecular weight PAHs, and BaP surpassed the threshold levels in practically the entire area. PAH-parent diagrams revealed a relatively homogenous fingerprint of four clusters obtained in a multivariate statistical study. In addition, molecular diagnostic ratios pointed to coal combustion as the main pollution source, whereas only some outliers appeared to be related to specific spills. A BaP threshold was calculated to be 0.24 mg kg-1, over 10 times the limit established in Spain. Finally, a factor analysis revealed a positive correlation of BaP with elements usually emitted in coal combustion processes, such as Tl and V. This observation fosters the hypothesis of a historical and indelible pollution fingerprint in soils whose sources, characteristics and potential environmental and health concerns deserve further attention. All things considered, caution should be taken when using soil screening levels in regions associated with coal exploitation and heavy industry.Aptamers are small, functional single-stranded DNA or RNA oligonucleotides that bind to their targets with high affinity and specificity. Experimentally, aptamers are selected by the systematic evolution of ligands by exponential enrichment (SELEX) method. Here, we have used rational drug designing and bioinformatics methods to design the aptamers, which involves three different steps. First, finding a probable aptamer-binding site, and second, designing the recognition and structural parts of the aptamers by generating a virtual library of sequences, selection of specific sequence via molecular docking, molecular dynamics (MD) simulation, binding energy calculations, and finally evaluating the experimental affinity. Following this strategy, a 16-mer DNA aptamer was designed for Annexin A1 (ANXA1). In a direct binding assay, DNA1 aptamer bound to the ANXA1 with dissociation constants value of 83 nM. Flow cytometry and fluorescence microscopy results also showed that DNA1 aptamer binds specifically to A549, HepG2, U-87 MG cancer cells that overexpress ANXA1 protein, but not to MCF7 and L-02, which are ANXA1 negative cells. We further developed a novel system by conjugating DNA1 aptamer with doxorubicin and its efficacy was studied by cellular uptake and cell viability assay. Also, anti-tumor analysis showed that conjugation of doxorubicin with aptamer significantly enhances targeted therapy against tumors while minimizing overall adverse effects on mice health.CRISPR-Cas9-mediated gene knockout and base-editing-associated induction of STOP codons (iSTOP) have been widely used to exterminate the function of a coding gene, while they have been reported to exhibit side effects. In this study, we propose a novel and practical alternative method referred to as CRISPR Start-Loss (CRISPR-SL), which eliminates gene expression by utilizing both adenine base editors (ABEs) and cytidine base editors (CBEs) to disrupt the initiation codon (ATG). CRISPR-SL has been verified to be a feasible strategy on the cellular and embryonic levels (mean editing efficiencies up to 30.67% and 73.50%, respectively) and in two rabbit models mimicking Otc deficiency (Otc gene) and long hair economic traits (Fgf5 gene).
