Field investigations by NIOSH suggest that the levels of respirable crystalline silica at well sites can exceed the permissible exposure limits. Thus, from an occupational safety perspective, it is important to evaluate the potential toxicological effects of FSD, including any neurological risks. Here, we report that acute inhalation exposure of rats to one FSD, i.e., FSD 8, elicited neuroinflammation, altered the expression of blood brain barrier-related markers, and caused glial changes in the olfactory bulb, hippocampus and cerebellum. An intriguing observation was the persistent reduction of synaptophysin 1 and synaptotagmin 1 proteins in the cerebellum, indicative of synaptic disruption and/or injury.     Tinospora cordifolia (TC) is being used as a blood purifier in Ayurveda since ancient time. It is a very popular immunomodulator and holds anti-inflammatory and anti-oxidative potential, hence anti-aging properties. Therefore, it is also known as ‘Amrita’ in Ayurveda and is widely used to treat diabetes mellitus type II (T2DM) and its secondary complications; however, its underlying mechanism was not expedited to date. AIM- To explore the in vivo therapeutic efficiency and mechanism of action of TC and its secondary constitute magnoflorine on the skeletal muscle atrophy in the rat model of T2DM.

    Animal model of T2DM was developed using streptozotocin (STZ) injection followed by intervention with TC, metformin, and magnoflorine for three weeks. Confirmation of T2DM and abrogation of atrophic markers and possible mechanisms on supplementation of TC and magnoflorine were explored using histology, bio-assays, Western blotting, and q-PCR.

    TC and Magnoflorine supplementations significantly (p≤0.05) decreased muscle, expression of IL-6, and slow MyHC indicated the increased myogenesis in response to magnoflorine and independent from its hypoglycemic activity.

    Investigate the effects of VSP on treating kidney infection induced by uropathogenic Escherichia coli (UPEC) and the underlying mechanisms.

    Both in vivo and in vitro infection models were established with the UPEC strain CFT073. After oral administration of VSP, the levels of bacterial load, cathelicidin (CRAMP), Toll-like receptors (TLRs) in the kidney were evaluated. The expression of cathelicidin (LL-37) in human renal cell carcinoma cell line (A498) was tested after the treatment of VSP.

    In the kidneys of infection models, high-titer bacteria was detected. In the kidney of rat model, the expression of CRAMP was down-regulated, no significant change was observed in the levels of TLRs. After oral administration of VSP, the bacterial load was significantly decreased in rat and mouse models, and the levels of CRAMP and TLRs were significantly up-regulated in rat model. In vitro, the expression of LL-37 was significantly inhibited by CFT073. VSP up-regulated the expression of LL-37 in A498cells.

    Borneol was widely used in traditional Chinese medicine formulas due to its pharmacological activities, e.g. sedative, anti-inflammatory, and anti-ischemic properties. Cinnamomum camphora (L.) J.Presl essential oil (BEO) is a by-product of natural crystalline borneol (NCB) production obtained by steam distillation of Cinnamomum camphora (L.) J.Presl leaves, and borneol was the main component of BEO. This study aims to investigate the anti-inflammatory effect of BEO and its corresponding mechanisms through in vitro and in vivo studies.

    Human erythrocyte membrane stability assay and the acute inflammation murine model (xylene-induced ear edema) were chosen to evaluate the anti-inflammatory effect of BEO. Expression of inflammatory mediators, including interleukin (IL)-1β, IL-6, and tumor necrosis factor α (TNF-α) was determined by real-time quantitative polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assays (ELISA). The functional compounds in the BEO were identified by using gas chromatography-mass spectrometry (GC-MS).
