Behavioural development is an important aspect of research on animal behaviour. In bats, many studies have been conducted on the development of flight behaviour, but the postnatal behavioural development of bats remains largely unexplored. We studied the behaviours and postnatal development of infant bats by conducting controlled video recorded experiments. Our results showed that before weaning, Asian parti-coloured bats (Vespertilio sinensis) were able to exhibit four types of behaviours, namely, crawling, head moving, wing flapping, and wing spreading, and these behaviours are different from those observed in experiments with adult bats. The number of occurrences of these behaviours was correlated with age and scaled mass index. Furthermore, the number of occurrences of these behaviours in young bats could also reflect their physical developmental status. In young bats, wing flapping and spreading might be a type of play behaviour. These behaviours were negatively correlated with the time of the first flight, indicating that they might help to promote individual physical development. Our results provide fundamental data for revealing the ontogenetic and neurophysiological mechanisms of behavioural development in bats.Danon disease is a severe X-linked disorder caused by deficiency of the lysosome-associated membrane protein-2 (LAMP-2). Clinical manifestations are phenotypically diverse and consist of hypertrophic and dilated cardiomyopathies, skeletal myopathy, retinopathy, and intellectual dysfunction. Here, we investigated the metabolic landscape of Danon disease by applying a multi-omics approach and combined structural and functional readouts provided by Raman and atomic force microscopy. Using these tools, Danon patient-derived cardiac tissue, primary fibroblasts, and human induced pluripotent stem cells differentiated into cardiomyocytes (hiPSC-CMs) were analyzed. Metabolic profiling indicated LAMP-2 deficiency promoted a switch toward glycolysis accompanied by rerouting of tryptophan metabolism. Cardiomyocytes’ energetic balance and NAD+/NADH ratio appeared to be maintained despite mitochondrial aging. In turn, metabolic adaption was accompanied by a senescence-associated signature. Similarly, Danon fibroblasts appeared more stress prone and less biomechanically compliant. Overall, shaping of both morphology and metabolism contributed to the loss of cardiac biomechanical competence that characterizes the clinical progression of Danon disease.Glucosinolates are a group of plant secondary metabolites that can be hydrolyzed into a variety of breakdown products such as isothiocyanates, thiocyanates, and nitriles. These breakdown products can facilitate plant defense and function as attractants to natural enemies of insect pests. As part of the diet, some of these compounds have shown cancer-preventing activities, and the levels of these metabolites in the edible parts of the plants are of interest. Selleck NSC 74859 In this study, we systematically examined variations in glucosinolates, their precursors, and their breakdown products in 12 commonly consumed vegetables of the Brassicaceae family with gas chromatography-quadrupole time-of-flight mass spectrometer (GC-Q-TOF/MS), liquid chromatography-quadrupole time-of-flight mass spectrometer (LC-Q-TOF/MS), and liquid chromatography-triple quadrupole mass spectrometer (LC-QQQ/MS), using both untargeted and targeted approaches. The findings were integrated with data from literature to provide a comprehensive map of pathways for biosynthesis of glucosinolates and isothiocyanates. The levels of precursor glucosinolates are found to correlate well with their downstream breakdown products. Further, the types and abundances of glucosinolates among different genera are significantly different, and these data allow the classification of plants based on morphological taxonomy. Further validation on three genera, which are grown underground, in damp soil, and above ground, suggests that each genus has its specific biosynthetic pathways and that there are variations in some common glucosinolate biosynthesis pathways. Our methods and results provide a good starting point for further investigations into specific aspects of glucosinolate metabolism in the Brassica vegetables.Adult studies show that healthy diet patterns relate to better sleep. However, evidence during adolescence, when sleep may change dramatically, is lacking. Within a cohort of 458 Mexican adolescents, we examined whether consumption of three dietary patterns was associated with sleep duration and timing measured 2 years later, as well as changes in sleep timing and duration. Dietary patterns (identified a posteriori in a prior analysis) were assessed with a baseline food frequency questionnaire, and sleep was measured with wrist actigraphy at baseline and follow-up. Linear regression analyses adjusting for sex, age, screen time, and smoking were conducted. Adolescents with higher consumption of a Plant-Based and Lean Proteins pattern had earlier sleep timing (-0.45 h with 95% Confidence Interval (CI) -0.81, -0.08 in the highest compared to lowest quartiles), less of a phase delay in sleep timing over follow-up (-0.39 h with 95% CI -0.80, 0.02), and shorter weekend sleep duration (0.5 h with 95% CI -0.88, -0.1). Higher consumption of an Eggs, Milk and Refined Grain pattern was associated with earlier sleep timing (-0.40 h with 95% CI -0.77, -0.04), while consumption of a Meat and Starchy pattern was related to higher social jetlag (weekend-weekday sleep timing difference). Healthier diet patterns may promote better sleep in adolescents.

Acute myeloid leukemia (AML) is one of the most common malignant and aggressive hematologic tumors, and its pathogenesis is associated with abnormal post-transcriptional regulation. Unbalanced competitive endogenous RNA (ceRNA) promotes tumorigenesis and progression, and greatly contributes to tumor risk classification and prognosis. However, the comprehensive analysis of the circular RNA (circRNA)-long non-coding RNA (lncRNA)-miRNA-mRNA ceRNA network in the prognosis of AML is still rarely reported.

We obtained transcriptome data of AML and normal samples from The Cancer Genome Atlas (TCGA), Genotype-tissue Expression (GTEx), and Gene Expression Omnibus (GEO) databases, and identified differentially expressed (DE) mRNAs, lncRNAs, and circRNAs. Then, the targeting relationships among lncRNA-miRNA, circRNA-miRNA, and miRNA-mRNA were predicted, and the survival related hub mRNAs were further screened by univariate and multivariate Cox proportional hazard regression. Finally, the AML prognostic circRNA-lncRNA-miRNA-mRNA ceRNA regulatory network was established.
